dc.description.abstractIn the last years, the reports of polymyxins resistance have increased considerably, so efficient, reliable, fast and easily interpreted testing to determine polymyxins susceptibility is necessary. The present study aimed to develop and evaluate the performance of a colorimetric microelution method for detecting polymyxin B resistance in clinical isolates of Gram-negative bacilli. The tests were performed with 11 clinical isolates, including 8 E. coli and 3 K. pneumoniae. The colorimetric polymyxin B microelution method was an adaptation of the colistin broth disk elution test, adding resazurin at the end of the process. The results were interpreted using the breakpoints of the BrCAST. When evaluating the microelution results with and without resazurin, 7 isolates presented categorical agreement and 9 and 10 isolates, respectively, presented essential agreement with the reference method. In conclusion, both polymyxin B microelution test and the colorimetric polymyxin B microelution test were easy to perform, requiring materials commonly found in routine microbiology laboratories. Furthermore, a method involving polymyxin B, which is the polymyxin used in Brazil, encourages the study of new susceptibility tests against polymyxins.pt_BR
dc.description.resumoNos últimos anos a resistência as polimixinas têm aumentado consideravelmente, portanto é de extrema importância métodos eficientes, confiáveis, rápidos e de fácil interpretação para determinar a suscetibilidade às polimixinas. O presente estuda objetivou desenvolver e avaliar o desempenho de um método de microeluição colorimétrico para detecção da resistência à polimixina B em isolados clínicos de bacilos Gram-negativos. Foram testadas 11 isolados clínicos, sendo 8 E. coli e 3 K. pneumoniae. O método colorimétrico de microeluição da polimixina B foi uma adaptação do método da eluição da colistina, adicionando resazurina no final do processo. Os resultados foram interpretados de acordo com os pontos de corte estabelecidos para polimixina B pelo BrCAST. Quando avaliado os resultados da microeluição com e sem a resazurina, 7 isolados estavam em concordância categórica e 9 e 10 isolados, respectivamente, em concordância essencial com o método de referência. Diante do exposto, podemos concluir que tanto o teste da microeluição como o teste colorimétrico de microeluição da polimixina B demonstraram facilidade de realização, além de requerer materiais comumente encontrados em laboratórios de microbiologia de rotina. Ademais, um método envolvendo a polimixina B, a qual é a polimixina utilizada no Brasil, vem fomentar o estudo de novos testes de suscetibilidade frente às polimixinas.pt_BR
